Project: This project aimed to develop a PMCA test designed to detect PrPTSE in blood from vCJD patients
Main results and related published data: This project was given financial support in 2014, and is still ongoing. However, some results have been recently published by the consortium in a paper showing that the development of a PMCA assay allows the detection of PrPTSE in only a few µl of blood samples, with not less than 100% sensitivity and specificity for vCJD patients. This success was due not onlyto an automatization of the method but also to the processing of the blood samples to enrich in PrPTSE and removed other blood componentsthatcould interfere with PMCA. Thiscould be achieved bycentrifugation of the blood sample in the presence ofsarkosyl, followed by a washing step in phosphate-buffered saline. The material was then used to seed PMCA reaction in the presence of brain homogenatesfrom transgenic mice overexpressing the human PrP gene (Met/Met 129).
